I am not sure whether we need TRY here but we don't need THE before TWO otherwise it is okay to say as you have written. Or you can say,

Don't create a dispute between two of us.

I agree with Willy that "the two of" is not necessary because the word "between" implies that there are only two people involved. If there were three or more people involved in the conflict we would want to use the word "among".

I think that there is a slight meaning difference between "Don't try to create a conflict between us" and "Don't create a conflict between us" but both sentences are grammatically correct.
